Abstract:

Background This study aimed to investigate whether Artificial Seminiferous Tubules (AST) coated with testicular extra cellular matrix (ECM) support maturation of spermatogenic cells from immature testes. MaterialsAndMethods Medical grade tubes coated with testicular ECM were used. Testes from immature mice were loaded in the concentration of 10000- 20000/ml. Tubes were then immersed in germ cell culture medium supplemented with a mixture of growth factors and cultured at 32oC for 30 days. After 7 days in culture maturation inducing factors including FSH, SCF and Retinoic Acid were added. After 14 days in culture, the contents of some tubes were collected and samples were taken for histology, DNA content and gene expression analyses. Also some cells were co-cultured with the Sertoli cell monolayer. After 3-4 weeks, cells with the morphology of spermatids and sperm were collected and their ability to fertilize eggs was determined by ICSI and IVF. Furthermore, the ability of the fertilized eggs to develop to embryos was investigated. Results After 7 and 14 days, small cells similar to the morphology of round and elongated spermatids were present. Localization of the Peanut Agglutinin (PNA) as well as PAS staining revealed the presence of acrosomal structure in these cells. Cells matured in AST expressed meiotic and post meiotic markers and on average 75% of the cells reached to haploid stage after two weeks of maturation. After 18 days in culture more elongated spermatids and cells with the appearance of mature sperm containing head, acrosome, mid piece and tail were formed. Injection of round spermatids developed in AST into MII eggs resulted in GFP embryos. Conclusion AST coated with testes ECM created a micro milieu similar to testicular seminiferous tubules and supported maturation of spermatogenic cells.

Localization of the DAZ gene expression in seminiferous tubules of patients with spermatogenic disorders.

The research on the expression and mutations of DAZ and its homologues in human and other mammals suggests that protein products of these genes can mainly affect development of germinal cells. The aim of the present study was to analyze the expression of the DAZ gene in seminiferous tubules of six men with spermatogenic disorders (hypospermatogenesis and spermatogenic arrest). The results based...

P-98: Ultrastructural Study of Basement Membrane of Seminiferous Tubules in Adult Diabetic Rats with Spermatogenic Alterations

Background: The complications of diabetes are the major problems that occur in diabetic patients. It has been reported that, insulin insufficiency and the impairment of regulatory action of this hormone on Leydig and Sertoli cells have an important role in testicular alterations. Testicular disorders and change in normal spermatogenesis is one of the consequences of diabetes. This study was des...

Histomorphometric evaluation of treatment of rat azoosper-mic seminiferous tubules by allotransplantation of bone marrow-derived mesenchymal stem cells

Objective(s): Bone marrow-derived mesenchymal stem cells (BM-MSCs) potentials make them appropriate for cell therapy including ability of differentiation and release of anti-inflammatory cytokines and growth factors secreta. For treatment of azoospermia to induce proliferation and differentiation of germ cells, MSCs transplantation has been introduced. The aim of the present experimental case-c...

Differentiation of human umbilical cord mesenchymal stem cells into germ-like cells in mouse seminiferous tubules

Our previous study demonstrated that human umbilical cord mesenchymal stem cells (HUMSCs) were capable of differentiation into germ cells in vitro. To assess this potential in vivo, HUMSCs were microinjected into the lumen of seminiferous tubules of immunocompetent mice, which were treated with busulfan to destroy endogenous spermatogenesis. Bromodeoxyuridine labeling studies demonstrated that ...

Protective Effects of Curcumin on the Structural Parameters of Seminiferous Tubules and Leydig Cells in Methandienone Treated Mice

Background and objectives: Although anabolic androgenic steroids (AAS) use has become became more prominent in bodybuilders and professional athletes, their consumption is nowadays a problem that involves a wider population. Methandienone or dianabol is an anabolic androgenic steroid which is used by athletes for fitness. This study was conducted to investigate, the likely prot...

A Note on the Morphology O F the Seminiferous Tubules of Birds

In the course of a study of the renal tubules of birds, by means of the maceration method devised by Huber,l in which full grown roosters (Gallus domestica) were used as material, the injection of the 75 per cent solution of hydrochloric acid was through the aorta central to the branches supplying the kidneys and sex glands. In a number of the cases the testes were found to be well injected and...
